چکیده

Targeting the inner nuclear membrane roteins destined for the inner nuclear membrane (INM) start out in the peripheral ER. Diffusion from the ER will get them to the contiguous outer nuclear membrane (ONM), but the next step could involve either vesicular transport, short-lived fusions between INM and ONM, or movement along the lipid bilayers surrounding nuclear pores. On page 1051, Ohba et al. suggest that the last explanation applies. Moreover, their data suggest that the process can only occur because the nuclear pore complex, which was thought to be a static structure, is constitutively remodeled. Previous models suggested that INM proteins move through the nuclear pore membrane, but researchers needed a dynamic assay to test the model. By adapting a protein trapping system (Chen et al. 1995. Proc. Natl. Acad . Sci. USA . 92: 4947–4951), Ohba et al. were able to watch fluorescently-labeled FRB reporter proteins move between the peripheral ER and the INM. Under normal conditions, the reporter moved between membranes without restriction. However, when the team added rapamycin to the cells, the FRB reporter accumulated in the INM as the reporter bound to an FK binding protein (FKBP) associated with the nuclear lamina. Thus, the rapamycin-mediated interaction trapped the reporter protein in the INM. The team thinks that native INM proteins become similarly trapped when they associate with nuclear structures. Only proteins whose luminal and cytosolic domains were under 60 kD gained access to the INM, a limitation noted previously. INM localization was dependent on both energy and temperature, which would be consistent with membrane fusion events. However, because the addition of inhibitors of membrane fusion had no effect on localization in the INM, the team hypothesizes that the energy is required for nuclear pore remodeling. Indeed, addition of antibodies against the nuclear pore protein gp210 blocked localization to the INM. While the pore structure is busy with the remodeling, small integral membrane proteins may slip by, passing into the INM. The team thinks pore remodeling is constitutive because reporter proteins that lacked any native nuclear protein sequence accumulated in the INM in the presence of rapamycin, suggesting that a signal is not required. Pathologies cardiac discrimination using the Fast Fourir Transform (FFT) The short time Fourier transforms (STFT) and the Wigner distribution (WD)

This paper is concerned with a synthesis study of the fast Fourier transform (FFT), the short time Fourier transform (STFT and the Wigner distribution (WD) in analysing the phonocardiogram signal (PCG) or heart cardiac sounds.     The FFT (Fast Fourier Transform) can provide a basic understanding of the frequency contents of the heart sounds. The STFT is obtained by calculating the Fourier tran...

The acoustic mechanics of stick slip friction in the California spiny lobster (Panulirus interruptus).

The dynamic interplay between static and sliding friction is fundamental to many animal movements. One interesting example of stick-slip friction is found in the sound-producing apparatus of many spiny lobster species (Palinuridae). The acoustic movements of the spiny lobster's plectrum over the file are generated by stick-slip friction between the two surfaces. We examined the microscopic anat...

Quantification of Parkinson Tremor Intensity Based On EMG Signal Analysis Using Fast Orthogonal Search Algorithm

The tremor injury is one of the common symptoms of Parkinson's disease. The patients suffering from Parkinson's disease have difficulty in controlling their movements owing to tremor. The intensity of the disease can be determined through specifying the range of intensity values of involuntary tremor in Parkinson patients. The level of disease in patients is determined through an empirical rang...

The effect of inversion times on the minimum signal intensity of the contrast agent concentration using inversion recovery t1-weighted fast imaging sequence

Background :Inversion recovery (IR) pulse sequences can generate T1-weighted images with a different range of inversion time (TI) to suppress or null the signal intensity (SI) for a specified tissue. In this study, we aimed to investigate the effect of TI values on the concentration of the contrast agent, which leads to a minimum signal intensity, using an inversion recovery T1-weighted 3-dim...

Modeling and Simulation of Substrate Noise in Mixed-Signal Circuits Applied to a Special VCO

The mixed-signal circuits with both analog and digital blocks on a single chip have wide applications in communication and RF circuits. Integrating these two blocks can cause serious problems especially in applications requiring fast digital circuits and high performance analog blocks. Fast switching in digital blocks generates a noise which can be introduced to analog circuits by the common su...

Strong dynamical effects during stick-slip adhesive peeling.

We consider the classical problem of the stick-slip dynamics observed when peeling a roller adhesive tape at a constant velocity. From fast imaging recordings, we extract the dependence of the stick and slip phase durations on the imposed peeling velocity and peeled ribbon length. Predictions of Maugis and Barquins [in Adhesion 12, edited by K. W. Allen, Elsevier ASP, London, 1988, pp. 205-222]...
